Activity 7: DNA Extraction

Purpose:

In order for students to understand how UV affects living organisms, they will benefit from actually extracting and handling DNA. This inexpensive lab reinforces discussions in the areas of enzymes, cell structure, lipid membranes, supercoiling, denaturation, solubility, viscosity, detergent/lipid interaction, macromolecules (polymers) and the nature of the bacterial genome.

Before beginning this activity. Students need to understand the importance of sterile technique

Materials:

For entire class
  • incubator (optional)
  • hot water bath (hot plate, large pot or beaker, thermometer)
  • Plates with E.coli colonies (1 plate for every 2 lab teams)

For each lab team

  • 1 eyedropper
  • 1 sterile inoculating loop
  • 6 ml of ethanol (95%) (keep on ice)
  • 2-15 ml culture tubes containing 4ml of Luria broth or some
  • suitable nutrient broth
